| - Introdukce genů rezistence k padlí travnímu obsažených ve dvou vzorcích Hordeum v. spontaneum do H. vulgare byla provedena křížením s odrůdou Tiffany. Testy rezistence na rostlinách F2 generace ukázaly poměr štěpení 15 : 1 u Tiffany x PI354949 a 3 : 1 u Tiffany x PI466495, což ukázalo na přítomnost dvou dominantních genů rezistence v prvním případě a jednoho dominantního genu ve druhém. K identifikaci jednotlivých R genů byla použita technologie mikrosatelitních markerů DNA a analýza rekombinace vazeb. Jeden z genů rezistence v PI354949 byl zjištěn v lokusu Mla ve vazbě s Bmac0213 (distální pozice 25,0 cM) a druhý na chromozomu 7H ve vazbě s Bmag0507 (proximální pozice 14,0 cM). U PI466495 byl gen R ve vazbě s Bmac0213 (distální pozice 8,4 cM) a je také lokalizován v lokusu Mla. Cílem této práce bylo jemné mapování perspektivního genu lokalizovaného na chromozomu 7H a identifikace těsně spojených markerů DNA pro výběr pomocí molekulárních markerů. (cs)
- The introduction of resistance genes to powdery mildew present in two accessions of Hordeum v. spontaneum into H. vulgare was performed by the cross with the variety Tiffany. Resistance tests on plants of F2 generation revealed the segregation ratio 15 1 in Tiffany x PI354949 and 3 : 1 in Tiffany x PI466495, which indicated presence of two and one dominant resistance genes, respectively. To identify individual R genes, technology of microsatellite DNA markers and linkage recombination analysis was applied. One of resistance genes in PI354949 was determined to be located in Mla locus linked with Bmac0213 (25.0 cM in distal position) and the other on chromosome 7H linked with Bmag0507 (14.0 cM in proximal position). In PI466495, R gene was linked with Bmac0213 (8.4 cM in distal position) and it is also located in Mla locus. The aim of this work is fine-mapping of the promising gene located on 7H chromosome and identification of tightly linked DNA markers for marker assisted selection.
