  • Během procesu přípravy a skladování trombocytárního koncentrátu je ovlivňována struktura a funkce trombocytu, dochází k apoptóze a aktivaci krevní destičky. Byl hodnocen stav buněčného poškození a aktivace trombocytů ze separátorů Haemonetics MCS+ a Trima Accel s odlišnou technologií aferézy. V den přípravy a exspirace byly vyšetřeny parametry buněčného poškození a aktivace trombocytu (pH, LDH, laktát, glukosa, annexin V, solubilní P-selektin). V den přípravy známky nejnižší aktivace vykázaly trombocyty ze separátoru Haemonetics MCS+. Na konci exspirace přípravky z aferézy vykázaly vyšší stupeň aktivace ve srovnání s trombocyty z plné krve, přípravky z obou separátorů byly srovnatelné. Na základě zhodnocení vývoje a vzájemných vztahů uvedených markerů, zejména P-selektinu a annexinu V se autoři domnívají, že počáteční aktivace trombocytů v přípravcích je pravděpodobně reversibilní. Je způsobena vlivem technologie přípravy a neovlivňuje proces apoptózy trombocytu během skladování. (cs)
  • During the preparation and storage of platelet concentrate the platelet structure and function are affected. It undergoes apoptosis and activation. The platelet storage lesion and activation were evaluated by means of separators Haemonetics MCS+ and Trima Accel with different technology of apheresis. Markers of platelet lesion and activation (pH, LDH, lactate, glucose,annexin V, sP-selectin) were evaluated at day of storage 0 and 5. The lowest degree of platelet activation was found in products obtained by Haemonetics MCS+ apparatus at the time of collection. Platelet concentrates obtained by apheresis revealed higher rise of LDH, annexin V and sP-selectin compared to buffy-coat derived platelets. On the basis of changes of sP-selectin and annexin V levels it could be concluded that initial platelet activation, which is induced by apheresis, is very likely without any further impact on quality of platelets during storage
