| - In our study, the ornithine (speC) and lysine (cadA) decarboxylase genes were investigated in E. coli strains using the PCR methods. 30 E. coli strains isolated from raw cow, sheep and goat bulk milk samples and cheeses obtained from different farms were evaluated. The alignment of the multiple sequences and analysis of conserved domains in the studied genes was carried out using ClustalW and BLAST programs. Analysis of the sequence sources for the speC gene revealed existence of speC isosyme, featured with homology between the 66–98 % compared to the speC nucleotide alignment. Also, in the case of cadA gene, we observed two groups of sequences with the homology of approximately 60 % (GenBank, cadA1, cadA2). On the basement of these facts, we designed and performed also additional PCR tests. 77 % strains were positive for speC, 73 % for speC isozyme gene, 60 % for cadA1 and 53 % for cadA2 PCR test. - V prezentovanej práci boli s pomocou metód PCR investigované gény ornitín (speC) a lyzín (cadA) dekarboxylázy v skupine izolátov E. coli. Testovanú skupinu kmeňov predstavovalo 30 kmeňov E. coli izolovaných zo surového kravského, ovčieho a kozieho mlieka a syra, vzorky pochádzali z viacerých fariem. Na porovnanie sekvencií skúmaných génov a na analýzu konzervovaných domén a štruktúry proteínov boli použité programy ClustalW a BLAST. Analýza dostupných sekvencií génu speC odhalila existenciu génu pre speC izozým, ktorého nukleotidové usporiadanie vykazovalo homológiu v intervale 66–98% v porovnaní so sekvenciou speC. Rovnako v prípade génu cadA sme zaznamenali existenciu dvoch skupín sekvencií s homológiou približne 60 % (GenBank, cadA1, cadA2). Na základe týchto faktov sme navrhli a optimalizovali doplnkové PCR testy. 77% kmeňov bolo pozitívnych na speC, 73% na prítomnosť génu speC izozým, 60% na cadA1 a 53% na cadA2 PCR test. Produkcia jednotlivých amínov v testovaných vzorkách bola úspešne overená.
