  • Cílem této práce bylo prozkoumat vztahy mezi různými metodami stanovení KMM za použití specificky modifikovaných standardních vzorků na mléčném základě s absencí některých rozpustných složek jako laktózy. Po prvním vyhodnocení byly do dalšího diskvalifikovány výsledky dvou metod, BI a FT-MIR pro velký posun a rozptyl hodnot, nevyhovující recovery a paralyzovaný vztah k ostatním metodám. Protože druhá metoda BI v hodnocení setrvala, jednalo se při diskvalifikaci pravděpodobně o lokální defekt v provedení. Postup vykazoval slabší recovery a potřebu metodické úpravy podporující věrohodnost výsledků jako zvýšení počtu kalibračních bodů oproti současné metodice. Výsledky metody FT-MIR byly silně systematicky posunuty v důsledku absence zejména laktózy . Korelace k výsledkům dalších relevantních metod byly těsné . Fotometrická metoda s Ehrlichovým činidlem (EH) vykázala přijatelné hodnoty všech hodnocených ukazatelů věrohodnosti. Nejpříznivějších výsledků dosahovala specifická metoda Ureakvant . (cs)
  • The goal of this work was to search the relationships between different methods of MUC determination with use of specifically modified samples on milk basis with absence of dissolved components such as lactose. The two method results, BI and FT-MIR were disqualified for large shift and variance of values, unsatisfacted recovery and paralysed relation to other methods. Therefore the second BI method was retained in the evaluation, it was probably a local deffect in the performance at disqualification. Procedure showed poorer recovery and necessity for methodical modifications for support of result reliability such as increase of number of calibration points as compared to contemporary procedure. The results of FT-MIR method were strong systematically displaced due to lactose absence in particular.The photometrical method with Ehrlich´s agent (EH) showed acceptable values of all the evaluated indicators of reliability. The specific Ureakvant method (UR) showed the most proper results.
