Description
  • De novo regeneration of pea tissue cultures has been demostrated to be a source of variation. On the other hand, pea regeneration via proliferation of organized meristems guarantee tru-to-type multiplication, at leats in short term culture. We have tested using molecular tools such as microsatellites and retrotransposon based DN markers, as well as ploidy assessment via flow-cytometry, the stability of 21- years old organogenic pe culture. No deviation from original were detected by all methods used as well as morphological deviation of obtained regenerants.

  • S využitím organogenní kontinuální in vitro kultury hrachu, založené před 21 lety, bylo pomocí molekulárních markerů a flow-cytometrie stdována genetické stabilita ve srovnání s původním osivem. byla konstatována stabilita jak na úrovni ploidie, tak rovněž na úrovni retrotransposonových a mikrosatelitních markerů.
