  • Poster:Current clinical management of pancreatic cancer is characterized by a lack of biomarkers useable in prediction of therapy response or determination of patients survival prognosis. Recent development in the area of epigenetic control of gene expression is bringing a new hope. Most importantly, with the discovery of RNA interference controlling mechanisms in the progression of pancreatic cancer, a whole family of microRNA biomarkers is finding its way though validation studies and into a clinical practice. While the majority of work is performed on archived pancreatic tissue originating from surgical resections, evaluation of bioptic tissue will likely be crucial in a clinical setting. Endoscopic ultrasound-guided fine needle aspiration biopsy (EUS-FNA) is currently routinely applied for acquisition of pancreatic tissue for cytology evaluations, however, utility of EUS-FNA samples for DNA and, in particular, miRNA testing is still rare. In this work we report on important experimental factors of endoscopic sampling and extraction of genetic material affecting the quality of DNA/miRNA testing from EUS-FNA samples. : Aside from optimization of laboratory protocols, factors of EUS-guided sampling have essential impact on the success rate of molecular tests. Proper selection of needle type and absence of aspiration are apparently key in minimizing blood contamination, high yield of extracted material with high portion coming from mutated (i.e. cancerous) cells.
