  • Alfalfa (Medicago sativa L.) is a forage legume of world-wide importance used in agriculture. The genetic relationship and distance among cultivars is of great interest for breeding program. Utilization of diverse germplasm is needed to enhance the genetic diversity of cultivars. Morphological traits and Simple Sequence Repeat (SSR) were used to evaluate genetic variability of 14 alfalfa cultivars in Czech Republic. Six SSR primers generated 27 polymorphic bands. The number of alleles per locus ranged from 2 for B21E13 to 6 for MTIC451 and MTIC189, with an average 4.5 alleles. The average polymorphic information content (PIC) was 0.67, ranging from 0.37 to 0.8. The lowest and the highest PIC value were recorded for primer B21E13 and MTIC189, respectively. The genetic diversity based on morphological characters ranged from 7.1 to 31.1. The highest genetic similarity (7.1) was found between Morava and Niva accession and the highest genetic dissimilarity (31.1) was found between Palava and Kamila items. The Jaccard´s coefficient based on molecular data varies from 0.21 (high genomic dissimilarity) to 1 (full genomic similarity), thus demonstrating the high polymorphism of the analyzed genotypes. The lowest genetic discance was recorded between Morava and Niva populations and highest genetic distance was calculated between Morava and Jitka & Niva and Jitka population. Cluster analysis based on morphological traits separated the accessions into 2 main groups and based on SSR data items formed into 2 main clusters with population Jitka comprising a singular group and 2 sub-clusters. We performed correlation analysis between the morphological and genetic diversity of 14 Medicago sativa populations. The matrices were compared using Pearson´s correlation coefficient (r = 0.159), which was very low. No significant correlation was detected between observed patterns of morphological and molecular variation.
