  • Tato práce se zabývala problematikou konzervace genetických zdrojů bramboru a chmele metodou kryoprezervace. Cílem této práce bylo uchování vybraných odrůd bramboru českého původu a významných genotypů chmele jako bezpečnostní zálohu v tekutém dusíku při teplotě -196 °C. Držitelé kolekcí – Výzkumný ústav bramborářský Havlíčkův Brod a Chmelařský institut Žatec zajišťují multiplikaci in vitro rostlin, hodnocení zdravotního stavu a ověřují identitu vybraných genotypů. Metody kryoprezervace jsou prováděny a vzorky uloženy ve Výzkumném ústavu rostlinné výroby, v.v.i. Praha. Je využívána jednoduchá kryoprezervační metoda předkultivace-dehydratace, která je založená na předkultivaci rostlin na mediu s vysokým obsahem přirozeného kryoprotektantu, sacharosy, následné dehydrataci vzrostných vrcholů nad silikagelem a jejich rychlém zamražení a odtání. U chmele předkultivace zahrnuje působení kryoprotektantu a nízké teploty. Předkultivace i dehydratace v suchém vzduchu způsobují snížení poklesu hydratace pletiv. (cs)
  • This work has dealt with the problem of potato and hop genetic resources conservation by the method of cryopreservation. Goal of this work is to store selected potato cultivars of the Czech origin and important hop genotypes as a safe duplicate in liquid nitrogen at temperature of -196 °C. The keepers of the crop germplasms - Potato Research Institute in Havlickuv Brod and Hop Research Institute in Zatec provide multiplication of in vitro plants and evaluate health state and genotype identity of selected genotypes. The cryopreservation methods and cryostorage of plant material are performed in the Crop Research Institute in Prague. A simple preculture-dehydration cryopreservation method used is based on in vitro plant cultivation on a medium with high level of a natural cryoprotectant, sucrose, subsequent dehydration of apical shoot tips above silicagel and ultrarapid rate of their freezing and thawing.
