| - Citlivá a rychlá metoda byla vyvinuta pro souběžné stanovení kyseliny methylmalonové, celkového homocysteinu a příbuzných metabolitů kobalaminového a folátového metabolismu v séru kombinací plynové chromatografie s hmotnostní detekcí. Thioly v disulfidických a proteinových vazbách byly vyvázány působením 2,3-dimerkaptopropansulfonové kyseliny a derivatizovány heptafluorbutylchlorformiátem v jediném kroku. Nové redukční činidlo poskytlo podstatně čistší extrakty než při použití dithiothreitolu. Nový postup umožnil GC-MS analyzovat sérové metabolity, včetně cystathionu a SH-dipeptidů. Přesná kvantifikace 7 biomarkerů byla umožněna nasazením deuterovaných kalibrátorů. Detekční limit byl v oblasti 7-20 nmol/l, inter-assay přesnost 1,5-8,8% a zpětný výtěžek se pohyboval v rozmězí 83-103%. Metoda kombinované redukce a derivatizace se ukázala jako účinná pro diagnostiku a skríning sérových metabolitů (cs)
- A sensitive and rapid method has been developed for simultaneous determination of serum methylmalonic acid, total homocysteine and related metabolites of cobalamin and folate status by gas chromatography-mass spectrometry. Thiols bound in proteins and disulfides were released with 2,3-dimercaptopropanesulfonic acid, and after deproteinization, they were derivatized with heptafluorobutyl chloroformate in a single step. The novel reducing agent afforded much cleaner extracts than those using dithiothreitol. The new method enabled GC-MS screening of serum aminoacidic metabolites, including cystathionine and thiol-containing dipeptides. Accurate quantitation of seven biomarkers was accomplished by using deuterated internal standards; the detection limits ranged from 7 to 20 nmol/L, the between-day precision from 1.5 to 8.8%, and the recoveries were between 83 and 103%.
