  • Environmentální rastrovací elektronový mikroskop (EREM) umožňuje pozorovat vzorky, které je obtížné či nemožné pozorovat v REM. Tlak v komoře vzorku v rozsahu od 1Pa do 2000Pa dovoluje pozorovat izolační vzorky bez výskytu nábojových artefaktů na jejich povrchu a vzorky obsahující vodu. Nicméně určité problémy přetrvávají při pozorování vysoce vlhkých biologických vzorků v jejich přirozené podobě a to především v průvěhu prvotních fází čerpání mikroskopu, kdy dochází k odpařování vody ze vzorků a jejichh poškození. Cílem této práce je najít a vytvořit takové metody, umožňující snízit odpar vody ze vzorku v EREM a zachovat tak jeho přirozenou podobu. (cs)
  • Environmental scanning electron microscope (ESEM) allows the visualisation of specimens that are difficult or impossible to image in a conventional high vacuum SEM. Pressure in the specimen chamber ranges from 1 Pa over 2000 Pa, which permits to observe specimens of beeing free from charging artefacts and containing different volume of water. But there are some difficulties with the observation of highly wet biological specimens in their natural state because during a pre-pumping process of the specimenn chamber an escape of water from the specimen occurs and thus its surface is damaged. The aim of this contribution is to find a method making possible to decrease water escape from the specimen during the initiation pumping process of the ESEM, immediately after the inserting of the specimen into the specimen chamber and during an image recording.
