Description
| - Cíl. Vyvinout HPLC metodu pro diagnostiku porfyrií v rámci laboratorního cvičení. Metoda. Vyvinuli jsme metodu pro stanovení koproporfyrinů I a III v moči pomocí kapalinové chromatografie s fluorescenční detekcí. Pro odseparování vybraných porfyrinů od interferujících látek jsme použili gradientovou eluci. Průtok mobilní fáze byl udržován na hodnotě 0,5 ml/min. Koproporfyriny I a III byly detekovány při těchto vlnových délkách: excitace: 394 nm a emise 624 nm. Výsledky. Analytické parametry jsou pro klinické testy dostačující. Variační koeficienty přesnosti v sérii byly pro koproporfyrin je 4,84% a koproporfyrin III 6,00%. Kalibrační křivky byly v rozmezí 6-460 ug/l (koproporfyrin I) a 6-490 ug/l (koproporfyrin III) lineární. Výtěžnost metody byla pro koproporfyrin I 92,23% (CV 4,38%) a koproporfyrin III 99,05% (CV 5,41%). Předběžné referenční hodnoty u skupiny dárců byly v rozmezí 973 +/- 220 ug/mol kreatininu (koproporfyrin I) a 1842 +/- 888 ug/mol kreatininu (koproporfyrin III). Závěr. Tato meto (cs)
- Objective. To develop a HPLC method for the diagnosis of porphyrias within the laboratory training. Method. We have developed and evaluated a method for the determination of coproporphyrins I and III in urine by high-performance liquid chromatography with fluorescence detection. By using the controller, a gradient program was used to separate selected porphyrins from other interfering compounds. The flow rate was 0.5 mL/min. Coproporphyrins I and III were detected at excitation 394 nm and emission 624 nm. Results. The analytical performance of the method is satisfactory for clinical trials. The intra-assay coefficients of variation were 4.84% and 6.00% for coproporphyrin I and III, respectively. The calibration curve was linear with the tested range 6-460 ug/L for coproporphyrin I and 6-490 ug/L for coproporphyrin III.
