| - Background. Catecholamines are metabolized by monoamino oxidase and aldehyd dehydrogenase to vanillylmandelic acid (VMA) and homovanillic acid (HVA). The major metabolite of serotonin is 5-hydroxyindoleacetic acid (5-HIAA). A number of diseases are accompanied by increasing or decreased catecholamine metabolism. Methods. This method is based on selective isolation of VMA, HVA and 5-HIAA from urine samples on AG 1-X8 resin. Separation of VMA, HVA and 5-HIAA was achieved on reversed phase HPLC with isocratic elution and coulometric electrochemical detection (EG = 450 mV; E1 = 180 mV, gain range = 200 nA; E2 = 400 mV, gain range = 200 nA). Results. The analytical performance of this method is satisfactory with intra-assay CV 4.8% for VMA, CV 6.5% for HVA and 2.8% for 5-HIAA, inter-assay CV 8.2% for VMA, CV 10.2% for HVA and 8.4% for 5-HIAA, recovery 92.3% for VMA, 90.1% for HVA and 97.6% for 5-HIAA. - Úvod. Katecholaminy jsou metabolizovány pomocí monoaminooxidasy a aldehyddehydrogenasy na kyselinu vanilmandlovou (VMK) a homovanilovou (HVK). Hlavním metabolitem odbourávání serotoninu je kyselina 5-hydroxyindoloctová (5-HIOK). Existuje celá řada onemocnění doprovázena buď zvýšenou nebo sníženou úrovní metabolismu katecholaminů. Metody. Metoda je založena na selektivní izolaci VMK, HVKa 5-HIOK z moče pomocí separace tuhou fází (AG 1-X8 pryskyřice). V dalším kroku jsou hladiny VMK, HVK a 5-HIOK v moči stanoveny pomocí RP-HPLC s coulometrickou elektrochemickou detekcí (EG = 450 mV; E1 = 180 mV, výstup 200 nA; E2 = 400 mV, výstup 200 nA). Výsledky. Analytické parametry prezentované metody jsou uspokojivé. Přesnost v sérii, vyjádřená variačním koeficientem, je pro VMK 4,8%, pro HVK 6,5% a pro 5-HIOK 2,8%. Přesnost mezi sériemi činí pro VMK 8,2%, HVK 10,2% a 8,4% pro 5-HIOK. Výtěžnost metody je pro VMK 92,3%, pro HVK 92,3% a pro 5-HIOK 97,6%.
