  • Vitamín D a jeho metabolity jsou důležité pro regulaci homeostázy vápníku a kostního metabolismu. Nejdůležitějšími metabolity vitamínu D jsou 25-hydroxyvitamín D3 a 1,25-dihydroxavitamín D3. Stanovení 25-hydroxyvitamínu D3 bylo provedeno pomocí vysoce účinné kapalinové chromatografie s UV detekcí. Pro separaci jsme zvolili jako stacionární fázi chromatografickou kolonu LiChroCart 125-4 Purospher Rp-18e, 5 um, mobilní fází byla směs methanolu a deionizované vody (95:5, v/v). Touto metodou jsme stanovovali hladiny 25-hydoxyvitamínu D3 u skupiny dobrovolných dárců krve. Předběžné hodnoty činí u této skupiny 10,42-116,20 nmol/l. Prováděli jsme statistickou analýzu porovnávající skupiny dárců se suplementací vitamínem D a bez suplementace. Žádné statisticky významné rozdíly jsme nenalezli. Vyvinuli jsme jednoduchou a selektivní HPLC metodu s UV detekcí pro stanovení 25-hydroxyvitamínu D3 v séru. Tato metoda poskytuje výbornou citlivost, přesnost a správnost a je vhodná pro výzkum, ale také pro klinické (cs)
  • Vitamin D and its metabolites are important for the regulation of calcium homeostasis and for bone metabolism. The most important metabolites of vitamin D are 25-hydroxyvitamin D3 and 1,25-dihydroxyvitamin D3. The determination of 25-hydroxyvitamin D3 was made by high-performance liquid chromatography with ultraviolet detection. For the separation, reverse phase column LiChroCart 125-4 Purospher RP-18e, 5 um, was used. The mixture of methanol and deionized water (95:5, v/v) was used as mobile phase. This method was applied to determination the concentration of 25-hydroxyvitamin D3 in a group of blood donors. The preliminary reference ranges of 25-hydroxyvitamin D3 in a group of blood donors are 10.42-116.20 nmol/L. The statistic analysis of the data measured was processed, including comparing the values obtained from the blood donors with supplementation of vitamin D and the ones without the supplementation of vitamin D. No statistically significant data diversity was found out in the analysis. We
