  • Beta-karoten hraje v biologických systémech významnou úlohu. Chrání organismus proti účinku reaktivních sloučenin kyslíku a dusíku. Vyvinuli jsme metodu pro stanovení beta-karotenu v lidské plazmě. Pro separaci jsme vybrali jako stacionární fázi chromatografickou kolonu LiChroCart 125-4 Purospher RP-18e, 5 um, mobilní fází byla směs methanolu a ethanolu (75:25, v/v). Jako vnitřní standard jsme používali retinylpalmitát. Analytické parametry předkládané metody jsou uspokojivé, přesnost v sérii byla s variačním koeficientem pod 10% a výtěžnost metody se pohybovala v rozmezí 90-100%. Kalibrační křivka byla lineární v celém rozmezí (0,02-2,00) umol/l. Detekční limit byl 0,5 pmol/20 ul nástřik. Tato metoda je reprodukovatelná a zcela vhodná pro stanovení beta-karotenu v lidské plazmě. Poděkování. Tato práce byla podpořena grantem MSM 0021627502. (cs)

A method for determination of beta-carotene in human plasma has been developed and evaluated. Chromatography was accomplished using isocratic elution on LiChrocart 125-4 Purospher RP-18e, 5 um, column. The mixture of methanol and ethanol (75:25, v/v) was used as a mobile phase. As internal standard was chosen retinyl palmitate. The analytical performance of this method is satisfactory, both intra-assay and inter-assay were with coefficients of variation below 10%, recovery was between 90-110%. The calibration curve was linear in the whole range tested (0.02-2.00) umol/L. The detection limit was 0.5 pmol/20 ul inject.
