| - Alzheimerova choroba (AD)je chronické, nevyléčitelné neurodegenerativní onemocnění. Za jednu z hlavních příčin AD je považován nedostatek acetylcholinu. Tento deficit může být způsoben příliš vysokou koncentrací a/nebo aktivitou enzymů, které acetlcholin rozkládají. Nejúspěšnější terapeutická strategie pro léčbu AD je využití inhibitorů cholinesteráz. Cílem naší práce je najít nová potenciální léčiva AD. Acetylthiocholin (ATCH) je používán pro testování enzynmatické hydrolýzy acetylcholinu in vitro, protože mechanismus hydrolýzy je kvantitativně shodný a rekační průběh hydrolýzy ATCH může být sledován on-line dvěma nezávislými metodami: spectrofotometrickou (Ellmanova metoda; stanovení produktu reakce thiocholinu) a electrochemickou (pH-statová metoda; stanovení kyseliny octové jako produktu). elamnova metoda byla použita pro testování mechanismu neinhibované enzymatické hydrolýzy ATCH butyrylcholinesterázou (BCHE) a acetylcholinesterázou (ACHE). - Alzheimer's disease (AD) is a chronic, incurable, neurodegenerative disorder. As one of the most common reason for AD lack of acetylcholine is considered. This deficit can be caused by too high concentration and/or activity of enzymes which breakdown acetylcholine. The most successful therapeutic strategy for AD tratment is application of cholinesterase inhibitors. The ideal goal of our research is to find a new potential drug against the Alzheimer´s disease. Acetylthiocholine (ATCH) is used for testing of enzymatic hydrolysis of acetylcholine (ACH) in vitro, because the mechanism of the hydrolysis is qualitatively similar to ACH and its reaction course can be quantitatively measured on-line by two independent methods: spectrophotometric (the Ellman´s method; determination of product thiocholine) and electrochemical (pH-stat method; determination of the product acetic acid).
