  • Plant cells are able to undergo postembryonic de novo organogenesis, and thus plant organs or an entire plant body are regenerated from previously differentiated cells. The long known phenomenon of in vitro plant regeneration suggests the principal role of two phytohormones, cytokinin and auxin. The identity of newly formed organs was shown to be dependent on different concentration ratios of these two phytohormones. This work focuses on the influence of auxin/cytokinin interaction in the early stages of de novo organogenesis. We used different markers (members of the cytokinin signaling pathway, auxin accumulation and transport) in a hypocotyl experimental system. As experimental model we selected Arabidopsis mutant lines affected in cytokinin signaling: single and double mutants in cytokinin receptors AHK2, AHK3 and AHK4. Organogenic response of ahk mutant lines was compared with the wild type during the first five days of cultivation.
