  • The aim of this study was to explore the functional interaction of retinoic acid (RA)- and okadaic acid (OA)-driven pathways in v-Myb-transformed monoblasts BM2. We found that 10 nm OA induces adherency, cell cycle arrest, phagocytic activity, production of reactive oxygen species, and expression of vimentin in BM2 cells. These features that mark differentiation along monocyte/macrophage pathway are enhanced in BM2 cells upon simultaneous treatment with OA and RA. Interestingly, the 20 nm OA induces rather apoptosis than differentiation of BM2 cells. This pro-apopotic effect of OA was inhibited by RA. These results indicate that pro-differentiation and pro-apoptotic effects of OA on BM2 monoblasts are differently regulated by RA.

  • Cílem této práce bylo prozkoumat funkční interakce signálních drah řízených kyselinou retinovou (RA) a kyselinou okadaiovou (OA) v monoblastech transformovaných onkogenem v-myb. Zjistili jsme, že 10 nm OA indukuje adherenci, fagocytickou aktivitu, tvorbu reaktivních kyslíkových radikálů, expresi vimentinu a zastavuje buněčný cyklus monoblastů BM2. Tyto znaky, které jsou považovány za markery monocytové/makrofágové diferenciační dráhy, jsou ještě posíleny u buněk BM2 vystavných současnému působení OA a RA. Je zajímavé, že 20 nm OA indukuje spíše apoptózu než diferenciaci buněk BM2 a tento pro-apoptotický účinek OA byl inhibován RA. Z našich výsledků vyplývá, že RA odlišně zasahuje do signalizace OA, která řídí diferenciaci a apoptózu monoblastů BM2. (cs)
