  • Při studiu diferenciace monoblastů transformovaných onkogenem v-myb (BM2) jsme v diferencujících buňkách pozorovali nárůst hladiny neznámého proteinu, který jsme hmotnostní spektrometrií identifikovali jako cytoskeletární protein vimentin. Transkripčně-aktivačními testy jsme zjistili, že nárůst hladiny vimentinu v buňkách BM2 vystavených forbolovému esteru TPA, trichostatinu A (TSA) či okadaikové kyselině (OA) koreluje s nárůstem aktivity promotoru genu kódujícího vimentin. Dále jsme pozorovali, že transkripce vimentinu je indukovaná transkripčními faktory c-Myb, v-Myb, c-Jun, v-Jun a také aktivovanými jadernými receptory pro kyselinu retinovou. Pro zjištění významu vimentinu při indukované diferenciaci BM2 jsme použili strategii snížení exprese vimentinu pomocí siRNA. U klonů BM2 se sníženou hladinou vimentinu, které byly kultivovány za přítomnosti TPA, respektive TSA, jsme pozorovali výrazně sníženou schopnost tvorby makrofágových polykaryonů a sníženou schopnost fagocytózy.   • Differentiation BM2 monoblasts upregulate expression of vimentin, the protein involved in formation of cellular cytoskeleton. Transactivation assays revealed that increased level of vimentin in BM2 cells treated with TPA, TSA or acadaic acid correlates with increased activity of vimentin gene promoter. In addition, vimentin gene transription is inducible by c-Myb, v-Myb, c-Jun, v-Jun transription factors as well as by ligand-activated retinoic acid receptors. In order to evaluate the role of vimentin in differentiation of BM2 cells, we silenced vimentin gene expression using siRNA. In the BM2 clones with decreased level of vimentin treated with TPA or TSA we observed significatly reduced capability of phagocytosis and efficiency of formation of macrophage polykaryons. These results indicate that strictly regulated increase of vimentin expression is essential for fromation of fully active macrophage polykaryons during monocytic/macrophage differentiation pathway.
