  • Koaguláza negativní stafylokoky jsou běžnou součástí mikroflóry kůže i kožních žláz teplokrevných živočichů včetně člověka, ale mohou se též podílet na vzniku různých infekčních onemocnění, a to především u oslabených jedinců. Z humánního klinického materiálu jsou občas izolováni zástupci komplexu Staphylococcus sciuri, původně spojovaní pouze s výskytem u živočichů, případně v potravinách. Druh S. sciuri má význam i hlediska přítomnosti homologu genu mecA, považovaného za předchůdce genu pro rezistenci k meticilinu neseného kmeny MRSA. V naší studii jsme pomocí ribotypizace, PCR a biotypizace analyzovali 72 humánních izolátů komplexu S. sciuri spolu s 6 referenčními typovými kulturami. Ribotypizace byla prováděna restrikčním enzymem EcoRI a sondou komplementární s 16S a 23S rRNA, vyhodnocení ribotypů bylo pomocí programu GelCompar II. K biotypizaci byly použity soupravy API Staph a ID 32 Staph včetně některých klíčových konvečních testů a dále analýza profilu celobuněčných proteinů (1D SDS-PAGE). Bio (cs)
  • Members of the Staphylococcus sciuri species group are infrequently isolated from human clinical material. In our study a collection of 72 clinical isolates originating from Czech Republic and 6 reference type strains of S. sciuri complex were characterized by ribotyping and biotyping. All strains were ribotyped using EcoRI restriction enzyme and probe complementary to 16S and 23S rRNA. The relatedness of ribotypes was evaluated by using the Dice coefficient and clustered by UPGMA method. Biotyping was performed with the API Staph and ID 32 Staph kits supplemented by conventional tests key for this bacterial group and by whole-cell protein profiles analysis (1D SDS-PAGE). Biochemical identification of isolates from S. sciuri species group based on commercial kits was often uncertain due to variations observed in some tests and required additional testing. All isolates and type strains were clearly distinguished by ribotyping according to their specific ribotypes at the species or subspecies level. The
