  • Bakteriofág 812, zástupce čeledi Myoviridae, je virulentní polyvalentní fág s �irokým rozmezím hostitelů, zahrnujícím kmeny a druhy rodu Staphylococcus. Tyto vlastnosti jej činí vhodným kandidátem pro fágovou terapii stafylokokových infekcí. Tato práce byla zaměřena na srovnání genomu a proteomu standardního typu fága 812, jeho mutant v rozmezí hostitele a příbuzných fágů U16, SK311, 131 a Twort s cílem objastnit molekulární základ jejich rozdílného hostitelského rozmezí. Genomy uvedených fágů tvořené lineární dvouřetězcovou DNA o velikosti 145 kb byly srovnávány pomocí restrikční analýzy. Úseky vykazující u jednotlivých fágů rozdíly byly pak charakterizovány detailně. Pro účely proteomických studií byla pou�ita 1-D SDS-PAGE. Nalezené proteiny byly identifikovány metodou peptidového mapování s vyu�itím MALDI-TOF hmotnostní spektrometrie. Srovnáním fágových genomů byly zji�těny vedle nukleotidových substitucí té� rozsáhlej�í přestavby sekvencí DNA. Inzerce a delece o velikosti ~1 a� 3 kb byly nalezeny (cs)
  • Virulent and polyvalent staphylophage 812, a member of Myoviridae family, exhibits strong lytic activity on a broad host range of Staphylococcus species, thus being a suitable candidate for treatment of staphylococcal infections. Genome of this phage is represented by linear 145 kb dsDNA. Our study was focused on genome and proteome comparison of 812 wild-type phage, its host-range mutants and related phages U16, 131 and SK311 with the aim to explain molecular basis of differences in their host-range. The genomes of phages under study were compared by DNA restriction analysis, and the genomic regions showing differences in individual phages were characterized in details. The proteome analysis included separation of phage protein lysates by 1D SDS-PAGE with subsequent identification of proteins by peptide mapping using MALDI-TOF mass spectrometry. Comparison of the phage genomes revealed several rearrangements of DNA sequences as well as single nucleotide substitutions in different parts of the genomes
