  • Saccharides and various glycoconjugates play a significant role in many host-pathogen interactions. Pathogen lectins that specially recognize sugar components can be involved in primary recognition of host organism. The Ralstonia solanacearum bacterium, an important plant pathogen that causes high agricultural losses, produces a fucose-binding lectin RSL. During its purification, another protein that reveals sugar affinity has been co-purified. This protein (MW 20034) displays mannose binding ability and N-terminal sequence shows high similarity with a hypothetical protein RSp0569 from R. solanacearum (ATCC11696 strain) genome GMI1000 megaplasmid. Preliminary biochemical experiments confirm mannose-binding lectin properties of the protein. Hexagonal shaped crystals were grown using hanging drop vapor diffusion and the diffraction data at 1.9 A resolution were collected under cryogenic conditions (100K) on beamlines ID14-1, at ESRF, Grenoble, France.
