| - Roztroušená skleróza (RS) je chronické autoimunitní onemocnění CNS. Matrixmetalloproteinázy (MMP) hrají klíčovou roli v imunopatogenezi RS a významně přispívají k progresi zánětlivého procesu. Je známo 23 MMP, které hrají roli v rozvoji RS. Cílem studie bylo analyzovat aktivitu MMP-2 a TIMP-2 u RS a jejich vztah k formě onemocnění. Sérové hladiny MMP-2 a TIMP-2 byly analyzovány metodou ELISA u vzorku 50 pacientů s jistou RS (průměrný věk: 36+/-8 let, průměrné EDSS skóre 3.8+/-1.8), z toho 30 mělo relabující a 20 chronicko progresivní formu onemocnění. Kontrolní skupinou bylo 50 zdravých dobrovolníků (průměrný věk 36+/- 9 let). Průměrné sérové hladiny MMP-2 a TIMP-2 se významně nelišily od hladin zjištěných ve skupině nemocných s RS, avšak hladiny byly signifikantně vyšší v podskupině RS s chronicko-progresivním průběhem ve srovnání s podskupinou s relabujícím průběhem (p< 0.05), zřejmě jako ukazatelm destrukce nervové tkáně. MMP se ukázaly jako slibný marker formy RS. (cs)
- Multiple sclerosis (MS) is a chronic autoimmune disorder of the CNS. Matrixmetalloproteinases (MMPs) play a key role in the immunopathogenesis of MS and are significant contributors to the progression of the inflammatory process. There are 23 MMPs known to play a role in the MS disease process. The aim of this study was to analyse the activity of MMP-2 and TIMP-2 in multiple sclerosis and to assess their relationship to the form of the disease. There were 50 patients with definite multiple sclerosis (mean age: 36+/-8 years, mean EDSS score 3.8+/-1.8) studied. Serum levels of MMP-2 and TIMP-2 were quantified by ELISA method in 30 patients with relapsing-remitting form, 10 with definite secondary progressive and 10 with primary progressive form of the disease and compared with those of 50 healthy controls (mean age 36+/-9 years).
