  • Haloperidol je jedním z klinicky nejčastěji používaných ligandů sigma receptorů. Jedná se o psychotropní látku, užívanou především při léčbě závažných psychiatrických onemocnění (schizofrenie, delirium apod.). Po jeho aplikaci jsou často popisovány nežádoucí vedlejší účinky na kardiovaskulární systém - tachyarytmie, KES, palpitace, vysoký krevní tlak a často i náhlá srdeční smrt. V naší práci jsme zjišťovali účinek haloperidolu na sodíkový proud INa a draslíkové proudy, přechodný proud z buňky Ito, proud na konci 250ms-impulzu IK,end (převážně sestává ze zpožděného proudu z buňky IK) a proud IK1. Iontové proudy byly měřeny metodou vnuceného napětí (voltage clamp) na celé buňce (whole cell patch clamp), na enzymaticky izolovaných komorových srdečních buňkách potkana kmene Wistar za pokojové teploty. Haloperidol reverzibilně inhiboval amplitudy všech zkoumaných proudů a tato inhibice byla koncentraně závislá (pod vlivem 1 umol/l haloperidolu - 39% inhibice INa, 39% inhibice Ito a 14% inhibice IK,end; po (cs)

Thus, we have investigated the effects of haloperidol on the sodium current INa and potassium currents, the transient outward current Ito, the current at the end of 250ms-impulse IK,end (that is mainly composed of the delayed rectifier current IK) and the inward rectifier current IK1. Experiments were performed on enzymatically isolated rat ventricular cardiomyocytes by whole cell patch clamp technique at room temperature. Haloperidol inhibited reversibly and in concentration-dependent manner amplitudes of all tested ion currents with 39% inhibition of INa, 39% inhibition of Ito and 14% inhibition of IK,end in the presence of 1 ľmol/l and 95% inhibition of INa, 80% inhibition of Ito, 37% inhibition of IK,end and 29 % inhibition of IK1 in the presence of 10 ľmol/l haloperidol.
