  • Ligand sigma receptorů haloperidol je psychotropní látka vykazující závažné vedlejší účinky na kardiovaskulární systém, především komorové arytmie včetně torsades de pointes. Je známo, že haloperidol prodlužuje repolarizační fázi akčního potenciálu a efektivní refraktorní periodu. Účinek haloperidolu na repolarizační draslíkové proudy u kardiomyocytů nebyl ovšem doposud studován. Provedli jsme předběžnou analýzu účinku haloperidolu (1umol/l) na přechodný proud z buňky Ito, který hraje významnou úlohu v počáteční repolarizační fázi akčního napětí. Experimenty byly prováděny u isolovaných komorových kardiomyocytů potkana metodou whole cell patch clamp při pokojové teplotě. Haloperidol indukoval pokles amplitudy a urychlení apparent inaktivace Ito. Průměrná inhibice Ito - hodnocená jako změna časového integrálu -byla 31.0 +/- 2.5 %. Inhibice proudu stejně jako zrychlení apparent inaktivace Ito nebyly napěťově závislé. Křivka ustálené inaktivace byla signifikantně posunuta negativním směrem o 3.4 mV. Časo (cs)

As was documented, haloperidol prolongs the repolarization phase of action potential and the effective refractory period. However, the effect of haloperidol on repolarizing potassium currents in cardiomyocytes has not been studied so far. Thus, we performed a preliminary analysis of the effect of haloperidol (1ľmol/l) on the transient outward potassium current Ito that plays a significant role in the initial phase of action potential repolarization. The experiments were performed on enzymatically isolated rat ventricular cardiomyocytes using the whole cell patch clamp technique at room temperature. Haloperidol induced a decrease of amplitude and an acceleration of apparent inactivation of Ito. The averaged inhibition of Ito evaluated as a change of its time integral was 31.0 ą 2.5 %.
