  • The role of biomembranes in the chronic toxicity of environmentally occurring chromium acetate hydroxide was investigated by using primary human fibroplasts. Transport of chromium acetate hydroxide across the plasma membrane of the cell, and the effects of chromium (III) ions on the plasma membrane as well as othher intracellular membranes, were determined during six weeks of continous exposure by using atomic absorption spectrometry, observation of cell morphology, membrane integrity assays (for lactate dehydrogenase leakage and lysosomal membrane disruption), and mitochondrial assays (for mitochondrial dehydrogenase activity and mitochondrial transmembrane potential analysis). The type of cell death induced by long-term exposure was determined in terms of phosphatidylserine externalisation, caspase-3 activation, and chromatin fragmentation.

  • Byla zkoumána specifická role biomembrán při toxicitě vyvolané octanem hydroxidu chromitého na lidských kožních fibroblastech in vitro. Během 6 týdnů kontinuální expozice k octanu hydroxidu chromitému byl sledován transport a intracelulární distibuce Cr (III) iontů ve fibroblastech. Dále byla sledována buněčná morfologie, membránová integrita (LDH test) a mitochondriální aktivita (potenciál a aktivita dehydrogenáz) u exponovaných buněk. Typ buněčné smrti byl určen pomocí specifických markerů (kaspáza-3, chromatinová fragmentace). Octan hydroxid chromitý v koncentraci 100 mikroM se akumuloval v buňkách, postupně degradoval plazmatickou membránu a mitochondriální funkce. Výsledkem byla buněčná smrt, a to jak apoptóza tak nekróza. Antioxidant koenzym Q v koncentraci 10 mikroM částečně potlačoval toxické vlivu Cr (III) iontů. Tyto výsledky ukazují, že membrány lidských fibroblastů jsou významnými cíly chronické toxicity octanu hydroxidu chromitého a tento systém je vhodný pro studia toxicity kovů. (cs)
