  • Expozice k šestivaznému chromu vyvolává různé nežádoucí účinky jako hluboké kožní vředy a alergickou dermatitidu. Díky jeho komplikované chemii mechanismus účinku chromu není zcela objasněn. V této práci jsme zkoumali vliv chromanu draselného (1.5-45 mikroM) na buněčnou motilitu cytoskeletální organizaci, oxidativní stres, mitochondrální aktivitu a aktivaci apoptózy u lidských kožních fibroblastů během 24 hodin. Chroman draselný indukoval časově a koncentračně závislou reorganizaci cytoskeletu, buněčné smrštění a ztrátu motilní aktivity. Tyto změny byly doprovázeny snížením mitochondriálního potenciálu, zvýšením oxidačního stresu a uvolněním cytochromu c. U umírajících buněk byla potvrzena aktivace kaspázy 3. Tato výsledky naznačují, že šestivazný chrom poškozuje cytoskelet a mitochondrie v časných stádiích a změny v těchto cílech aktivují na mitochondriích závislou i nezávislou apoptózu. (cs)

Because of many potential intracellular targets for hexaval are not entirely understood. To investigate the role of the cytoskeleton and mitochondria in this process, primary human dermal fibroplasts were exposed to various concentrations of potassium chromate for 24 h. The followed markers included cell motility, cytoskeletal organization, oxidative stress, mitochondrial activity and activation of the apoptotic cascade. Potassium chromate (1.5-45 mikroM) induced time- and concentration-dependent cell shrinkage, reorganization of cytoskeleton and loss of motile activity in fibroplasts. In some cells this was followed by membrane blebbing. Dynamic changes in cell morphology were accompanied with the loss of mitochondrial membrane potential, increased oxidative stress and release of cytochrome c.
