| - We analysed porcine antigen-specific immunity six months after experimental infection with Mycobacterium avium subsp. avium (MAA) by measuring IFN-gamma production (ELISA, qRT-PCR and flow cytometry) and lymphocyte transformation test after in vitro re-stimulation with the MAA-derived antigen. In MAA-infected animals, IFN-gamma was increased at both mRNA and protein levels in response to the specific MAA antigen in the blood, spleen and mesenteric lymph nodes. Similarly, a positive antigen-driven response was detected by proliferation assay. Using flow cytometry, we found double-positive CD4+CD8+ lymphocytes as the major T lymphocyte subset producing IFN-gamma after specific re-stimulation. In conclusion, antigen-specific immune response in swine is retained for at least six months after infection with MAA. Furthermore, we identified the presence of antigen-specific double-positive CD4+CD8+ cells in MAA-infected pigs.
