  • In this work functional status in boar spermatozoa collected from frozen-thawed semen and capacitated in vitro by caffeine was studied. The motile spermatozoa of boars were isolated by gradient, treated with 1mM or 2mM caffeine and capacitated in IVF medium or co-incubated with matured porcine oocytes. Motility, viability and acrosome integrity or fertilizing ability of spermatozoa were evaluated. The acrosome reaction was faster for 1mM caffeine-treated compared with 2mM caffeine-treated spermatozoa in all sires but its progress was different in each boar. The penetration rate was higher for 2mM caffeine-treated compared with 1mM caffeine-treated spermatozoa. On the contrary, monospermy was higher and total fertilization was significantly higher for 1mM caffeine-treated compared with 2mM caffeine-treated spermatozoa for all tested boars. It can be concluded that the progress of acrosome reaction was specific for spermatozoa of each boar and influenced the efficiency of fertilization process.

  • V práci byl studován funkční status spermií kapacitovaných kofeinem získaných ze zmrazeného spermatu kanců. Motilní spermie byly izolovány na gradientu Percollu, ošetřeny 1mM or 2mM kofeinem a kapacitovány v IVF mediu nebo ko-inkubovány ze zralými prasečími oocyty po dobu tří hodin. Byla hodnocena jak motilita, viabilita a integrita akrozomu tak i fertilizační schopnost spermií. Akrozomální reakce byl rychlejší u spermií ošetřených 1mM kofeinem ve srovnání se spermiemi ošetřenými 2mM kofeinem u všech kanců ale rozvoj akrozomální reakce byl odlišný u každého z nich. Úroveň penetrace byla vyšší u spermií ošetřených 2mM kofeinem ve srovnání se spermiemi ošetřenými 1mM kofeinem. Naproti tomu monospermie byla vyšší a celková fertilizace byla signifikantně vyšší u spermií ošetřených 1mM kofeinem u všech testovaných kanců. Lze konstatovat, že rozvoj akrozomální reakce byl specifický pro spermie individuálních kanců a ovlivnil účinnost fertilizačního procesu. (cs)
