| - V této práci jsme analyzovali 23 terénních kmenů Salmonella enterica serovar Typhimurium (S. Typhimurium) na přítomnost konjugativních plazmidů přenášejících rezistence k antibiotikům. Konjugace a přenos rezistencí k antibiotikům byl pozorován u 12 kmenů. Ve dvou případech nebyla při konjugaci přenesena do recipientní buňky rezistence k tetracyklinu a ampcilin asi u 10% transkonjugantů. Bližší charakterizace těchto kmenů ukázala, že ztráta obou rezistencí současně souvisela se ztrátou transpozonů Tn3 (blaTEM) a Tn1721 (tet(A)) z konjugativních plazmidů p9046 a p9134. Pokud se však v průběhu konjugace ztrácela pouze rezistence k tetracyklinu, transpozon Tn1721 byl nahrazen sekvencí pocházející z plazmidu virulence S. Typhimurium obsahující část genu rck, a geny srgA, srgB, ORF7 a pefI . Tyto dva plazmidy byly tedy schopné rekombinace s plazmidem virulence S. Typhimurium, což vedlo k vytvoření plazmidu kódujícího jak geny pro rezistence k antibitiokům, tak geny ovlivňující virulenci salmonel. (cs)
- In this study we analysed field isolates of Salmonella enterica serovar Typhimurium (S. Typhimurium) for the presence of conjugative plasmids transferring resistances to antibiotics. Altogether 23 strains were analysed for the presence of conjugative R plasmids. Conjugation and transfer of antibiotic resistance was observed in 12 strains. In two cases, resistance to tetracycline and/or ampicillin was not transferred by conjugation to ~10% of transconjugants. Detailed characterisation showed that the loss of both resistances was associated with the loss of Tn3 (blaTEM) and Tn1721 (tet(A)). However when only the tetracycline resistance was lost, the Tn1721 was replaced with a partial sequence of rck, and coding sequences of srgA, srgB, ORF7 and pefI originating from the S. Typhimurium virulence plasmid. Two plasmids from our collection were capable of recombination with the virulence plasmid of S. Typhimurium and subsequently spread both antibiotic resistance and virulence genes to the recipient.
