| - Cílem studie bylo zjistit výskyt viru hepatitidy E (HEV) v českých chovech prasat pomocí nested RT-PCR. Vzhledem k vysoké variabilitě genomu HEV byly k tomuto účelu použity tři sady specifických primerů. Celkově bylo vyšetřeno 32 selat z 11 chovů. Tato selata byla indikována k vyšetření na syndrom celkového chřadnutí selat po odstavu (PMWS). Od každého zvířete byl odebrán vzorek žluči, jaterní tkáně a séra. Nejčastěji RNA HEV pozitivní byly vzorky žluči (42.9%), následovaly vzorky jaterní tkáně (16.1%) a sér (3.2%). Nejméně jedno RNA HEV pozitivní sele bylo detekováno v sedmi chovech (63.6%; celkový počet 11). Specifická 242 bp sekvence ORF1 části HEV genomu českých izolátů byla následně sekvenována a fylogeneticky charakterizována. Fylogenetická analýza potvrdila zařazení těchto izolátů HEV do genotypu III rodu Hepevirus. Porovnání získaných sekvencí se sekvencemi dostupnými v GenBank ukázalo naprostou jedinečnost českých HEV izolátů. (cs)
- The objective of the present study was to carry out a small surveillance programme in Czech pig production herds using the nested RT-PCR to trace hepatitis E virus (HEV) in different biological samples. Due to the high genetic variability of HEV, three sets of primers of its genome were used. A total of 32 piglets from 11 herds clinically suspected of postweaning multisystemic wasting syndrome (PMWS) were examined. Bile, liver and serum samples were collected from each animal. HEV RNA was most frequently detected in the bile samples (40.0%), followed by liver (16.1%) and serum (3.2%). Seven (63.6%) of the 11 monitored farms were found to have at least one HEV RNA positive piglet. Specific 242 bp sequences within the ORF1 were sequenced and phylogenetically analysed. Phylogenetic analysis confirmed that all detected Czech swine HEV (CZswHEV) isolates belonged to genotype III. Comparison of the CZswHEV isolates with sequences of swHEV available in GenBank failed to find any 100% homologous HEV isolate.
