  • Data z microarray analýz u Salmonella Typhimurium ukázaly, že největší variabilita mezi kmeny tohoto serovaru je založena na přítomnosti či nepřítomnosti profágů v genomu. To nás vedlo k myšlence využít přítomnosti profágů jako cíle pro PCR diferenciaci terénních kmenů. V této práci jsme proto optimalizovali 4 triplex PCR specifické pro 12 cílových sekvencí převážně fágového původu a u 102 kmenů jsme pak prokázali, že tato multiplex PCR má téměř stejnou rozlišovací schopnost jako pulsní gelová elektroforéza (22 různých PCR typů a 25 různých PFGE typů). Přestože měla PFGE mírně vyšší rozlišovací schopnost, vzhledem k jednoduchosti a snadnosti sdílení výsledků představuje multiplex PCR zajímavou alternativu k PFGE typizaci Salmonella Typhimurium.   • Recent data from microarray analysis showed that the most frequent sources of genomic variation between different Salmonella enterica serovar Typhimurium (S. Typhimurium) strains are integrated prophages. This led us to a hypothesis that PCR detection of the integrated prophages might be an efficient typing tool alternative to pulsed-field gel electrophoresis (PFGE). In this study we therefore optimised 4 triplex PCRs specific for 12 target sequences of mostly prophage origin and tested them in 102 field strains. The same set of strains was characterized also by PFGE. Among these strains, 22 different multiplex PCR and 25 different PFGE profiles were identified. Despite the fact that the PFGE was slightly more discriminatory, the multiplex PCR typing due to its simplicity and potential of simple data sharing between laboratories represents an interesting, user-friendly alternative to the PFGE typing of S. Typhimurium.
