  • The members of Mycobacterium avium species, comprising M. a. subsp. paratuberculosis (MAP), M. a. hominissuis (MAH), M. a. avium (MAA), M. a. silvaticum (MAS), are currently the most prevalent opportunistic pathogenic mycobacteria causing mycobacterial infection in animals and humans. The ability to distinguish between these subspecies is of relevance for proper diagnosis and control programmes of the diseases. The aim of this study was to design a fast and specific PCR strategy for the detection and differentiation of M. avium subsp. from the solid plate cultures for use in routine veterinary diagnosis. We have developed a multiplex PCR based on IS900, IS901, IS1245 and the dnaJ gene. This method allows the detection of MAP, MAH and MAA/ MAS in one PCR reaction and theoretically enables mixed infections of MAP and MAA or MAP and MAH to be revealed. The sensitivity of this multiplex PCR 1000 CFU enabled the use of this test directly for DNA isolated from the tissue of the heavily infected sheep.

  • Druh Mycobacterium avium, obsahující poddruhy M. a. subsp. paratuberculosis (MAP), M. a. hominissuis (MAH), M. a. avium (MAA), M. a. silvaticum (MAS), je v současné době jeden z nejvíce rozšířených oportunních patogenů infikujících jak zvířata tak lidi. Rozlišení jednotlivých zástupců je důležité jak z hlediska správné diagnostiky, tak při přípravě preventivních programů. Cílem této studie bylo navrhnout rychlou a specifickou PCR pro detekci a rozlišení jednotlivých zástupců z již narostlých bakteriálních kultur. Proto jsme navrhli multiplex PCR, který využívá specifických inzerčních sekvencí IS900, IS901, IS1245 a genu dnaJ. Metoda tak umožňuje detekci MAP, MAH a MAA/ MAS v jedné PCR reakci a teoreticky umožňuje detekovat mix infekce MAP a MAA nebo MAP a MAH. Citlivost této metody je kolem 1000 CFU, což činí tuto metodou využitelnou též pro přímé detekce jednotlivých zástupců z masivně infikovaných tkání. V této studii jsem navrženou metodu použili na detekci MAP ze tkání ovcí. (cs)
