Description
| - Stable lyophilised preparation of paclitaxel (7 mol%) encapsulated in the liposomes was prepared by various procedures: hydration of lipid film, the proliposome-liposome method and lyophilisation from 2-methylbutan-2-ol. Extrusion through 0.2-um polycarbonate membranes was applied to reduce the size distribution of liposomes to about 180 - 190 nm (PDI 0.1). A negative charge (zeta-potential of -30 mV in saline phosphate buffer) was sufficient to prevent flocculation and fusion of liposomes in suspension during storage. An isocratic reverse-phase high performance liquid chromatography (HPLC) method with ultraviolet (UV) detection was used for determination of paclitaxel in liposomal preparations. The value of paclitaxel encapsulation efficiency (EE) was determined to be approximately 95%. Mannitol, lactose, sucrose and trehalose were tested as cryoprotective agents. Except for mannitol, all the tested saccharides were equally cryoprotective at lipid:saccharide molar ratios of 1:5 and 1:10.

- Stabilní lyofilizovaný liposomální preparát obsahující 7 mol % paclitaxelu byl připraven různými metodami: hydratací lipidového filmu, metodou proliposom-liposom a lyofilizací z 2-methylbutan-2-olu. Extruze přes 0,2 um polykarbonátové membrány byla použita pro snížení distribuce velikosti liposomů na 180 - 190 nm (PDI 0,1). Negativní náboj (hodnota zeta-potenciálu -30 mV v PBS pufru) úspěšně chránil liposomy před jejich fůzí a agregací během uchovávání. Vysokoúčinná kapalinová chromatografie (HPLC) s UV detekcí byla použita pro stanovení obsahu paclitaxelu v liposomálních preparátech. Hodnota enkapsulační účinnosti paclitaxelu (EE) byla stanovena na 95 %. Manitol, laktóza, sacharóza a trehalóza byly testovány jako kryoprotektiva. Všechny testované sacharidy, kromě manitolu, měly stejný kryoprotektivní účinek při molárním poměru lipid:sacharid 1:5 a 1:10. (cs)
