  • Robertsonian translocation rob(16;20) in the heterozygous state was discovered in a subfertile bull of the Czech Siemmental breed. The present experiments were designed to detect the rob(16;20) and determine its incidence for in vitro produced embryos(the 6- to 8-cell stage and the blastocyst stage), using fluorescence in situ hybridization (FISH) and the rob(1;29) as a detection control. To characterize semen of both bulls with the rob translocations, their sperm was examined for DNA integrity bythe Sperm Chromatin Structure Assay (SCSA). The embryos were fixed in methanol and acetic acid with Tween 20. Painting probes for chromosomes 16 (sp. Green) and 20 (sp. Orange) and chromosomes 1 (sp. Orange) and 29 (sp. Green) were simultaneouslyhybridized. Inthe embryos derived from the rob(16;20) bull, the presence of this translocation was not detected. On the other hand, 52.5 % of the embryos derived from the rob(1;29) bull were translocation carriers.

  • Robertsonovská translokace byla poprvé popsána u subfertilního býka českého simmentálského skotu. Práce si klade za cíl zjistit incidenci této translokace v in vitro produkovaných embryích (ve stadiu 6- 8 buněk a stadiu blastocysty) pomocí fluorescenčníin situ hybridizace (FISH). Jako kontrola je použita translokace rob(1;29). Mimo jiné byla provedena charakteristika integrity chromatinu ve spermiích těchto býků pomocí Sperm Chromatin Structure Assay (SCSA). Embrya byla fixována pomocí metanolu a kys.octové s přídavkem Tweenu 20. Byly použity malovací sondy pro chromosomy 16 (sp. Green) a 20 (sp. Orange), dále pro chromosomy 1 (sp. Orange) a 29 (sp. Green). V embryích pocházejících od býka s rob(16;20) nebyla nalezena fúze v žádném z embryí, zatímco52,5 % embryí pocházející od býka s rob (1;29) neslo translokaci. (cs)
