  • The aim of this study was to confirm whether the co-cultivation of Staphylococcus aureus and Streptococcus uberis with neutrophils of the bovine mammary gland leads to signs of apoptosis. In the study, 16 mammary glands of four virgin heifers aged 16 to18 months were examined. Apoptosis was detected by morphological features, by determination of histone-associated DNA fragments (ELISA), and by Annexin V and propidium iodide positivity (flow cytometry). S. aureus and S. uberis reduced the incidence ofkaryopyknotic and zeiotic neutrophils, and reduced the concentration of histone associated DNA fragments. The incubation of neutrophils with bacteria, however, increased the proportion of Annexin V positive cells and Annexin V and propidium iodidepositive cells. Co-cultivation of S. aureus and S. uberis with neutrophils led to the induction of phosphatidylserine translocation characteristic for the early stage of apoptosis.

  • Předmětem práce bylo zjistit, zda ko-kultivace Staphylococcus aureus a Streptococcus uberis s neutrofily mléčné žlázy skotu vede ke znakům apoptózy. Ve studii bylo použito 16 mléčných žláz 4 jalovic ve stáří 16-18 měsíců. Apoptóza byla detekována dlemorfologických znaků, determinací DNA fragmentů (ELISA) a pozitivitou na Annexi V a propidium iodid (průtoková cytometrie). Obě bakterie redukovaly výskyt karyopyknotických a zeiotických neutrofilů a koncentraci tzv. histone associated DNA fragmentů.Inkubace neutrofilů však vedla ke zvýšení podílu Annexin V pozitivních a Annexin V a propidium iodid pozitivních buněk. Ko-kultivace S. aureus a S. uberis s neutrofily vedla k translokaci fosfatidylserinu charakteristickou pro ranné stádium apoptózy. (cs)
