  • The objective of this study was to determine whether neutrophil apoptosis and their consequent elimination by macrophages from the mammary gland is modulated by an infection caused by Staphylococcus aureus (S. aureus). Instillation of S. aureus resultedin an intramammary infection which persisted during the following experimental period. The total number of both Annexin-V-positive and PI negative neutrophils and karyopyknotic neutrophils peaked at 24 h after both of PBS and S. aureus administration.Thehighest percentages of Annexin-V-positive and PI negative neutrophils and karyopyknotic neutrophils were detected 48 and 168 h after PBS and S. aureus administration, respectively. The total number of MPO+ macrophages was the highest 24 h and 48 hafter PBS and S. aureus administration, respectively; the percentage of MPO+ macrophages was the highest at 72 h in both cases. Mechanisms of cell pathogen elimination as well as inflammation resolution were very intensively involved.

  • Předmětem této studie bylo stanovit, zda apoptóza neutrofilů a jejich následná eliminace z mléčné žlázy makrofágy je modulována infekcí způsobenou Straphylococcus aureus (S. aureus). Intrammamární aplikace PBS a S. aureus vedla během 24 h k zánětlivéreakci, charakterizované nárůstem celkového počtu buněk a relativního zastoupení neutrofilů. Infekce S. aureus trvala po celou dobu pokusu (168 h). Celkový počet Annexin V pozitivních neutrofilů a karyopyknotických neutrofilů byl nejvyšší 24 h po PBS a168 hpo S. aureus. Po 24 h byl zaznamenán pokles celkového a relativního počtu neutrofilů. V obou případech dynamika odrážela zahájení rezoluce zánětlivé odpovědi. Procenticky nejvyšší podíl Annexin V positivních a karyopyknotických neutrofilů bylzaznamenán 48 h po PBS a 168 h po S. aureus. Celkový počet MPO+ makrofágů byl nejvyšší 24 h po PBS a 48 h po S. aureus, procento MPO+ makrofágů 72 h u obou. Modulační vliv infekce na průběh zánětu byl velmi slabý. (cs)
