Description
  • PALM microlaser technology was used to generate bovine, porcine and equine chromosome paint probes. Single chromosomes were isolated by laser dissection of the membrane around the target chromosome and 10-15 copies of the chromosome were collected by catapulting them into the Eppendorf cap. The microdissected DNA was amplified by degenerate oligonucleotide-primed PCR. The amplification product was labelled with digoxigenin and spectrum orange in secondary PCR, or by Biotin Chem-Link, and hybridized onto metaphase spreads. Fluorescence in situ hybridization signal revealed specific painting of the chromosome. Paint probes generated by this method are presented.
