  • Cílem bylo založit sbírku českých odrůd jiřinek a optimalizovat podmínky dlouhodobého uchování Dahlia pinnata v in vitro podmínkách. Bylo pozorováno, že zvýšená koncentrace sacharózy v médiu a nízká teplota měla pozitivní vliv na přežívání explantátů. Pro zachování české kolekce D. pinnata byla nutná detekce a eliminace DMV. Pro detekci DMV byla použita PCR metoda, pro získání zdravých rostlin z infikovaného materiálu byla použita meristémová kultura. (cs)
  • The aim of this work was to establish in vitro Czech dahlia collection and to investigate conditions of long-term conservation of Dahlia pinnata under in vitro conditions. It was observed that increasing concentration of sucrose in medium and low temperature had positive influence on survival explants. For conservation of the Czech collection of D. pinnata detection and elimination DMV were necessary. PCR (polymerase chain reaction) method was used for detection of DMV. The meristem culture was subsequently used for obtaining healthy plants from infected cultivars.
