  • The effect of selected in vitro and in vivo applied antimitotic agents on embryo production, morphology, on the rate of DH and abnormal plants was evaluated in four genotypes of Brassica carinata. In vivo colchicine application showed to be not efficient enough and unreliable, in the case of oryzalin the amount of abnormal plants was relatively high. Colchicine applied in vitro appeared to be one of the best chemicals in our experiments for its ability to increase the number of cotyledonary embryos, to speed up the development of maturing embryos and above all for its efficiency in chromosome doubling. In the case of trifluralin, the percentage of DH plants (86.2%) was the highest and stable among all successive replications and genotypes. With respect to above mentioned facts, trifluralin could be more efficient, cheaper and less toxic substitution of widely used antimitotic agent colchicine for production of doubled haploid lines also in cultivars and breeding materials of Brassica carinata.

  • Efekt vybraných in vivo a in vitro aplikovaných antimitotických látek na produkci embryí, morfologii, podíl DH a abnormálních rostlin byl stanoven u 5 genotypů B. carinata. In vivo aplikace kolchicinu se ukázala být neefektivní, v případě oryzalinu byl zjištěn relativně vysoký podíl abnormálních rostlin. In vitro aplikace kolchicinu se ukázala být jednou z nejefektivnějších metod z důvodu zvýšení embryogeneze, zrychlení vývoje embryí a především díky diploidizační efektivitě. Trifluralin zvýšil podíl DH rostlin na 86,2 % - tím se stal nejefektivnější látkou v našich experimentech, navíc výsledky byly stabilní u všech genotypů a opakování. Na základě těchto zjištění je možno konstatovat, že trifluralin může být účinnější, levnější a méně toxická náhrada běžně používaného kolchicinu k produkci dihaploidních linií také u kultivarů a šlechtitelských materiálů Brassica carinata.
