| - Příspěvek se zabývá možností využití akumulace proteinu dehydrin 5 (DHN5) jako markeru mrazuvzdornosti u ječmene. Mladé rostliny ječmene (21 kultivarů) patřící ke třem růstovým typům (přesívky, ozimy, jařiny) byly vystaveny otužení při neutrální fotoperiodě (12 h). Akumulace DHN5 byla stanovena densitometrickou analýzou imunoblotů a dosažená úroveň mrazuvzdornosti byla stanovena metodou přímých mrazových testů. Otužení vedlo k indukci akumulace DHN5 a k nárůstu mrazuvzdornosti u všech sledovaných kultivarů. V průběhu otužení se však projevily jak v akumulaci DHN5, tak v mrazuvzdornosti, rozdíly mezi odlišnými růstovými typy. Po třech týdnech otužení byla nalezena pozitivní lineární korelace mezi množstvím naakumulovaného proteinu DHN5 a dosaženou úrovní mrazuvzdornosti, avšak tato korelace byla dosažena jen tehdy, pokud byly hodnoceny kultivary s kontrastní mrazuvzdorností (ozimy a jařiny). - The article discusses the possibility of the use of dehydrin 5 protein (DHN5) accumulation as a selection marker for frost tolerance (FT) in barley. Young barley plants of 21 cultivars of three growth habits (intermediate, winter, spring) were exposed to cold acclimation (CA) under neutral (12 h) photoperiod. DHN5 accumulation was determined by immunoblots and the acquired level of FT was evaluated by direct frost tests. CA led to the induction of DHN5 accumulation and to the increase in FT in all cultivars examined; however, differences in the rate of DHN5 and FT increase between the growth habits occured. After three weeks of CA, a positive linear correlation between the amount of accumulated DHN5 and the acquired level of FT was found, but only when the cultivars with contrasting levels of FT (intermediate and winter cultivars versus spring ones) were used for evaluation.
