  • Efekt kolchicinu, trifluralinu a oryzalinu na depolymerizaci mikrotubulů a zdvojování chromozómů byl testován v embryogenních kulturách vybraných genotypů Brassica carinata. Přímá aplikace těchto mikrotubulárních inhibitorů vedla k obnově fertility dihaploidních rostlin. Aplikace 10 mikromolů/l roztoku trifluralinu během prvních 24 hodin kultivace byla vyhodnocena jako nejlepší pro zvýšení frekvence dihaploidních rostlin. Také všechny použité koncentrace oryzalinu a kolchicinu zvýšily významně podíl dihaploidních rostlin ve srovnání s neošetřenou kontrolou. (cs)
  • Microspore culture has been used to produce desirable meiotic recombinants in numerous Brassica species. However, the utilization of these recombinants relies on inefficient genome doubling procedures to generate fertile doubled haploids plants. Therefore, it represents a simple and rapid procedure to generate fertile doubled haploids in Brassica carinata. The effects of colchicine, trifluralin and oryzalin on microtubule depolymerization and chromosome doubling in embryogenic microspore cultures of Brassica carinata genotypes were examined. The direct application of these microtubule inhibitors to microspore cultures resulted in the recovery of fertile doubled haploids plants. The application of 10µM trifluralin during 24hrs of microspore culture was found to be the superior method for doubled haploid production. Similarly, all tested concentrations of colchicine and oryzalin significantly increased doubled haploid production in comparison with control variants.
