  • In the last decade, the safety of the collection of vegetatively propagated crops in the Czech Republic led also to the application of the cryopreservation technology. At present, the main vegetatively-propagated crops cryopreserved in the Czech cryobank include Allium spp., potato, apple, pear, Prunus spp., strawberry and hop. In apple, a “two-step freezing” protocol and the encapsulation-dehydration method are used with dormant buds and shoot tips, respectively. The “ultra-rapid cryopreservation method”, consisting in the quenching in liquid nitrogen of aluminium foils containing dehydrated shoot tips, is used for potato and hop meristems.   • Bezpečnost kolekcí vegetativně množených plodin vedlo v uplynulé dekádě k aplikaci kryoprezervace v České republice. V současnosti, hlavní vegetativně množené plodiny kryoprezervované v České kryobance zahrnují následující plodiny: Allium, brambor, jabloň, hrušeň, Prunus spp., jahodník a chmel. Pro kryoprezervaci jabloní je používán postup dvoustupňového mrznutí u dormantních pupenů a metoda enkapsulace-dehydratace u vzrostných vrcholů. Metoda ultra-rychlého mrznutí, která se využívá u bramboru a chmele, spočívá v rychlém zchlazení dehydratovaných vzrostných vrcholů na hliníkových plíšcích jejich rychlým ponořením do tekutého dusíku. U rodu Allium byly nejlepší výsledky dosaženy se vzrostnými vrcholy izolované z pacibulek a ošetřené roztokem PVS3 (50% glycerol, 50% sacharosa) přímo ponořenými do tekutého dusíku.
