Description
  • For identification of varieties of cereals we used new electrophoretic system. The Experion automated electrophoresis system applies a combination of microfluidic separation technology and sensitive fluorescent sample detection to perform rapid and automated analysis of protein. It automatically performs all steps of gel-based electrophoresis (sample separation, staining, destaining, imaging, band detection, and even some data analysis) to deliver reproducible separation and quantitation results in just 30 minutes. In our laboratory we used this system for analysis of protein of wheat (LMW and HMW glutenin subunits) and barley (hordeins). This approach was compared with the classic electrophoresis these proteins in polyacrylamide gel and sodium dodecyl sulphate. Results were characterized by considerable reproducibility and especially protein patterns of wheat obtained these techniques were very comparable.

  • V posledních letech byla vyvinuta tzv.chipová elektroforéza, která je miniaturizovanou modifikací kapilární elektroforézy. Chipová elektroforéza, představuje kombinaci mikrofluidní separační techniky a citlivé fluorescenční detekce pro kvantifikaci obsahové látky (DNA, RNA nebo proteinů). Dosud je použitelná pouze pro elektroforetické separace v přítomnosti dodecylsulfátu sodného (SDS). V naší laboratoři ve VÚRV byla úspěšně vyzkoušena pro analýzy frakce nízkomolekulárních (LMW-GS) a vysokomolekulárních (HMW-GS) podjednotek gluteninů pšenice a hordeinů ječmene – oba postupy klasické elektroforézy(SDS-PAGE) se používají pro identifikaci genotypů (odrůd, linií) i pro markerování některých znaků jakosti, které jsou s těmito proteinovými složkami ve vazbě. Metoda chipové elektroforézy byla porovnána s klasickou metodou elektroforézy (SDS-PAGE). Pomocí obou technik byly získány srovnatelné výsledky. Výsledky čipové elektroforézy se vyznačovaly vysokou reprodukovatelností. (cs)
