Description
| - Clavibacter michiganensis subsp. sepedonicus (further Cms), causing ring rot of potato, is a quarantine bacterium. Diagnosis of Cms is carried out by means of IFAS and bioassay on eggplant (EU directive No. 93/85/EEC), but PCR, ELISA and individual nutrient tests are also recommended. the latter could be substituted using BIOLOG (Biolog inc., USA) or fatty acid analysis (FAA). According to colony morphology, Cms occurs mostly as fluidal (smooth, mucoid), but sometime as intermediate (less fluidal) and nonfluidal (rough, nonmucoid) variants, too. In this study, commercial monoclonal antibodies (Agdia, USA), Pastrik primers PSA-1/PSA-R and dot blot hybridization with digoxigenin-labelled probe were used for determination of Cms strains representing all forms. All Cms strains were reliably identified by IFAS, although apart from typical also atypical cells were sometime recorded in populations of several rough strains.
