Description
  • Clavibacter michiganensis subsp. sepedonicus (Spieckman & Kotthoff) Davis et al. causes the serious ring rot disease of potato in many potato-growing areas around the world. Immunochemical diagnosis of the ring rot bacterium is based on the use of high-quality monoclonal antibodies among several serological methods, of which ELISA belongs to the most important. Although monoclonal antibodies are able to detect very low concentrations of the target bacterium, Cms usually is not found at concentration of 103 cells/ml or less. Therefore new techniques and ways are reached to enhance diagnosis of Cms. Commercial monoclonal antibodies (Agdia, USA) were used for determination and detection of Cms in potato tuber extracts by means of DAS-ELISA (double antibody sandwich enzyme-linked immunosorbent assay) and DASI-ELISA (double antibody sandwich indirect enzyme-linked immunosorbent assay), both with an enrichment step. Four semi-selective media, MTNA, NCP-88, PW 7 and PW 10 were used for cultivation of Cms, i
