| - Acaroid mites are serious pests of stored products and households. They produce allergens and transmit mycotoxin-producing fungi. The infestation of stored food by mites strongly decreases food safety and endangers public health. There is urgent need forrapid detection methods such as ELISA. The aim of our work was therefore to prepare and characterise antibodies for detection of the flour mite (Acarus síro). Two polyclonal antibodies (anti- A.siroK53 and anti-A.siroK54) were prepared against an antigen in the form of protein fraction from whole body homogenates of Acarus síro. Specificity and sensitivity of the prepared antibodies were evaluated. We did not observe any cross-reactivity with stored beetles, moths, other mite species, micro-fungi, therearing diet of mites (yeast and wheat diet) and extracts from wheat kernels. Cross-reactivity to Acarus gracilis was found out. This phenomenon is discussed in relation to the evolution position of this species.

- Roztoči ze skupiny Acaridida jsou závažnými škůdci ve skladovaných potravinách a domácnostech. Produkují alergeny a roznášení mikroskopické houby, produkující alergeny. Infestace skladovaných potravin roztoči výrazně snižuje bezpečnost potravin. To vyvolává nutnost existence rychlých detekčních metod jako je ELISA. Cílem práce bylo připravit a charakterizovat protilátky pro detekci roztoče Acarus siro. Byly připraveny dvě protilátky proti proteinové frakci z těl dospělců. Vlastnosti protilátek (jejich specifičnost a citlivost) byly hodnoceny a byl stanoven detekční limit pro detekci dospělců a jejich exkrementů. (cs)
