  • A total of 38 mucoid strains of Clavibacter michiganensis subsp. sepedonicus (Cms) were grown on culture media under different conditions e.g., of sugar concentration and temperatures with the aim to specify factors, which induce or stimulate a change of colony morphology from the mucoid wild type to nonmucoid form. It was found that stress conditions during cultivation, i. e. temperatures above 28°C (29-31°C), and culture media containing lower quantity of sugars, i.e. less than recommended level, stimulated the changes in colony morphology. The degree of mucoid growth of Cms strains decreased under the influence of these stress factors. Nevertheless, the influence of tested factors was not sufficient to change mucoid strains irreversibly to nonmucoid ones. Partially nonmucoid variants were termed %22intermediate%22 in colony morphology. Using DAS-ELISA and Biolog GP MicroPlate SystemTM, mucoid and intermediate variants of Cms were tested to see if these techniques might serve to identify both these va
