| - Cíle: naše pilotní studie si dala za cíl určit účinky jednonukleotidového promotorového polymorfizmu tumor-nekrotizujícího faktoru-alfa (TNF-alfa) 308 G?A v pediatrické zánětlivé chorobě střev (IBD), jeho vliv na zánětlivou aktivitu a klinickou manifestaci. Metody: genomická DNA od 164 subjektů (věk 8-18 let) - 82 s dlouhodobou IBD; 46 s Crohnovou chorobou (CD); 36 s ulcerativní kolitidou (UC); 82 zdravých kontrol byla genotypována pomocí testu restrikčních enzymů a polymorfizmus byl určen ve smyslu aktivity C-reaktivního proteinu a vlastní aktivity nemoci. Výsledky: byly nalezeny významné rozdíly v polymorfizmu (TNF-?) 308 G?A mezi IBD a kontrolami a UC a kontrolami. Frekvence TNF-alfa 308 A genotypu byla významně vyšší u pacientů s CD s penetrující chorobou v porovnání s pacienty bez komplikací a zdravými kontrolami. Distribuce alel a hladiny CRP byly nezávisle spojeny s CD komplikacemi. Závěry: TNF-alfa 308 A polymorfizmus může hrát úlohu při modifikaci CD fenotypu (cs)
- Our pilot study aimed to determine the effect of tumor necrosis factor-alpha (TNF-alfa) 308 G?A promoter single-nucleotide polymorphism in pediatric inflammatory bowel disease (IBD), its influence on inflammatory activity and the clinical manifestations. METHODS: We obtained genomic DNA from 164 subjects, 82 with long-standing IBD aged 8 to 18 years: 46 with Crohn disease (CD) and 36 with ulcerative colitis (UC). CONCLUSIONS: Although not necessarily dictating IBD initiation, the TNF-alfa 308 A polymorphism may play a role in modifying the CD phenotype. The polymorphism may influence disease activity as well as more intense inflammatory activity in both forms of IBD and may modify the progression of chronic digestive tract inflammation.
