  • Aggregation of meso-tetratolylporphyrins substituted with two (P2), three (P3) and four (P4) pyridinium groups and their complexation with DNA were studied using UV/Vis and fluorescence spectroscopies, resonance light scattering and circular dichroism. In aqueous solution porphyrins P2-P4 existed in a monomeric form and/or as H- and J-aggregates. The concentration ratio of monomer/aggregates was controlled by the number of cationic substituents, by ionic strength and by temperature. Porphyrin monomers were externally bound to calf thymus DNA with the binding constant of /~106 M-1. Porphyrin aggregates formed chiral assemblies on the DNA template not affecting the duplex melting point. The lifetime of the triplet states of bound monomeric P3 and P4 increased after binding, however it did not prevent the formation of 1O2.

  • Agregace, fotofyzikální vlastnosti a komplexace s DNA meso-tetratolyl porfyrinových derivátů substituované dvěmi (P2), třemi (P3) a čtyřmi (P4) pyridiovými skupinami byly studovány metodami UV/Vis spektroskopie, rozptylem světla, cirkulárním dichroismem a pomocí časově rozlišené luminiscencí singletního kyslíku. Ve vodném roztoku se P2 – P4 vyskytují jako monomery a agregáty obou typů (J i H). Poměr koncentrací monomeru a jednotlivých agregátů je řízen počtem kationických substituentů, iontovou silou a teplotou. P4 je převážně monomerní a váže se externě na DNA s telecího brzlíku s vazebnou konstantou 1.2x106 M-1. Pofyriny P2 a P3 samovolně agregují na fosfátové kostře DNA a tvoří chirální agregáty, které neovlivňují bod tání duplexu DNA. Doba života tripletních stavů monomerních P3 a P4 roste po navázání na DNA, ale tato skutečnost nebrání schopnosti tvořit singletní kyslík. (cs)
