Description
  • (i) Development and documentation of the potential of two alternative techniques employing time-of-flight mass spectrometry (TOF MS) to enable fast and comprehensive metabolite profiling in pome trees (ed parts and fruits). (ii) Investigation the possibility to distinguish the “healthy” and phytoplasma infected samples on the basis of the data generated by TOF MS (measurements of metabolom). (iii) Critical assessment of the novel instrumental approach with currently used strategies employing PCR and/or ELISA based techniques. Evaluation of the complementarily of TOF MS and bioassays as an effective tool in prediction / early detection of phytoplasma in pome fruit. (iv) Consideration of the potential of novel TOF MS strategies in characterization of other quality parameters of pome fruit varieties widely grown in Czech republic (Golden Delicious, Melrose, Idared). Implementation of LC-MS/MS for identification of quality markers. (en)
  • V rámci interdisciplinárního projektu bude zavedena nová strategie průkazu fytoplazmové infekce jabloní a zhodnocen její vliv na jakost plodů. Pro záznam změn metabolomických profilů bude využita hmotnostní spektrometrie s průletovým analyzátorem.

  • Methods employing liquid chromatography-mass spectrometry and novel ionization technique, direct analysis in real time (DART) were developed for metabolomics-based analysis of phytoplasma infected apple trees. Statistical analysis of obtained metabolomic fingerprints facilitated discrimination of infected and non-infected plants and selection of characteristic markers of phytoplasma infection. (en)
  • Pro metabolomickou analýzu vzorků jabloní napadených fytoplazmou byly vyvinuty metody využívající kapalinovou chromatografii s hmotnostní spektrometrii a novou ionizační techniku pro přímou analýzou v reálném čase (DART). Statistická analýza získaných metabolomických záznamů umožnila rozlišit infikované a zdravé rostliny a selektovat charakteristické markery onemocnění. (cs)
